PT-141 was discovered as an agent for sexual activity enhancement by accident. It owes its existence to Mac Hadley, a scientist who was performing experiments on his body. He experienced an eight-hour erection after administering a double dose Melanotan II, an early compound. A-MSH testing began in the 1960s with rats and continued into the 1980s. The peptide hormones Melanocortins regulate many physiological functions. Melanotan I, a Melanocortin analog was used as a tanning agent and Melanotan II for sexual dysfunction.

In 1960s research, PT-141 stimulated mPOA receptors in the brains and induced sexual arousal.
Many rounds of trials and research have been done on PT-141 over the years since the initial experiments. The effectiveness of subcutaneous delivery of 1.75mg of Bremelanotide versus placebo was tested.
